PD318088(PD-318088) is a non-ATP competitive allosteric MEK1/2 inhibitor. MEK1 and MEK2 are closely related, dual-specificity tyrosine/threonine protein kinases found in the Ras/Raf/MEK/ERK mitogen-activated protein kinase (MAPK) signaling pathway. The MEK1 inhibitor PD318088 binds simultaneously with ATP in a region of the kinase active site that is adjacent to the ATP-binding site. Birb796 binding to p38  extends into the ATP site, but also accesses this back pocket, partly overlapping the region where PD318088 binds in MEK1
